ABSTRACT: The purpose of this study was to develop and to assess the psychometric characteristics of a mobility scale for dogs. The original ten questions were reduced using validation process. One hundred and twenty three dog owners were invited to answer the questionnaire. Internal consistency, factor analysis, floor and ceiling effect and construct validity were studied. Good internal consistency (Cronbach's Alpha=0.854) was determined with two items eliminated. The instrument comprises 8 final questions, each of which has five possible answers (never, rarely, sometimes, often and always) scored between 0 and 4 or between 4 and 0 (for the items with inverse score). Three hypotheses proposed for the construct validity were verified: 1) gender does not influence dog mobility (P=0.584); 2) mobility decreases with age (P<0.001); 3) dogs with orthopaedic or neurological diagnosed pathologies have less mobility (median score (P25; P75) 46.9% (31.3; 68.8)) than healthy dogs (median score (P25; P75) 81.3% (71.9; 93.8)) (P<0.001). Total score range was 0 to 32 points, with higher values indicating greater mobility of dogs. The Dog Mobility Scale was capable of assessing mobility in dogs, with good psychometric characteristics, and is simple and inexpensive to apply in clinical practice.


RESUMO: Este estudo teve como objetivos construir e avaliar as caraterísticas psicométricas de uma escala de mobilidade para cães. O questionário, constituído inicialmente por dez questões, foi submetido a um processo de validação, recorrendo à redução do número de questões. Cento e vinte e três donos de cão foram convidados a responder ao questionário. Foram estudados a consistência interna, a análise fatorial, os efeitos teto e chão e a validade de construto do instrumento. Foi encontrada uma boa consistência interna (Cronbach's Alpha=0.854) com a eliminação de dois itens. O questionário/escala final ficou composto por oito questões, cada qual com cinco respostas possíveis (nunca, raramente, às vezes, frequentemente e sempre) pontuadas entre zero e quatro ou entre quatro e zero (para os itens com pontuação inversa). Três hipóteses propostas para a validade de construto foram verificadas: 1) o gênero não influencia a mobilidade (P=0.584); 2) a mobilidade declina com o envelhecimento (P<0.001); 3) cães com patologia ortopédica ou neurológica diagnosticada apresentam menor mobilidade (median score (P25; P75) 46.9% (31.3; 68.8)) do que cães saudáveis (median score (P25; P75) 81.3% (71.9; 93.8)) (P<0.001). A escala final resulta numa pontuação de 0 a 32 pontos em que maior pontuação corresponde à maior mobilidade. A Escala de Mobilidade do Cão avaliou, com boas caraterísticas psicométricas, a mobilidade de um grupo díspar de cães, revelando-se simples e econômica na sua aplicação na prática clínica.

The objective of the study described in this article was to characterize the antimicrobial resistance profiles among E. coli strains isolated from cohabitant pets and humans, evaluating the concurrent colonization of pets, owners, and home surfaces by bacteria carrying the same antimicrobial-resistant genes. The authors also intended to assess whether household surfaces and objects could contribute to the within-household antimicrobial-resistant gene diffusion between human and animal cohabitants. A total of 124 E. coli strains were isolated displaying 24 different phenotypic patterns with a remarkable percentage of multiresistant ones. The same resistance patterns were isolated from the dog's urine, mouth, the laundry floor, the refrigerator door, and the dog's food bowl. Some other multiresistant phenotypes, as long as resistant genes, were found repeatedly in different inhabitants and surfaces of the house. Direct, close contact between all the cohabitants and the touch of contaminated household surfaces and objects could be an explanation for these observations.

Recent studies of canine malignant mammary tumours suggest that reduction of E-cadherin and/or beta-catenin correlates with invasive behaviour and lymph node metastasis. The aims of this study were to examine the interrelationships between the expression of E-cadherin and beta-catenin, and the relationship between the expression of E-cadherin and/or beta-catenin and the mode of growth and metastatic capacity of canine malignant mammary tumours. 90 spontaneous malignant tumours and local and regional lymph nodes were studied. A significant relationship was evidenced between membranous expression of E-cadherin and beta-catenin (p=0.0027), but not between E-cadherin and cytoplasmic beta-catenin. Only E-cadherin as a separate factor was significantly related to tumour invasion (p=0.0072) and lymph node metastasis (p=0.0001). Neither membranous nor cytoplasmic beta-catenin expression was significantly related to either of these phenomena.

The relationship between MIB-1 labelling indices (LI), as detected by immunohistochemical methods, and other clinico-pathological characteristics was studied in a series of 77 malignant mammary tumours surgically removed from 47 female dogs. The immunostaining was assessed on the basis of the estimated percentage of positive cells in the areas of highest labelling. Multivariate logistic regression demonstrated no influence of breed, age, previous pregnancies, previous progestin administration, histological type or location of the tumour on MIB-1 LI. MIB-1 LI was significantly related to the size of the tumour, necrosis, invasive growth and histological grade, but not with ulceration, lymph node metastasis, skin fixation or E-cadherin expression. The significant relationship between MIB-1 LI and other known factors of poor prognosis suggests that a high LI may have prognostic value in canine malignant mammary tumours.

Cystinuria in Newfoundland dogs is a metabolic disease associated with a nonsense mutation in the exon 2 of the Slc3a1 gene. Similar to type I human cystinuria, heterozygote carriers are not affected by the disease and do not reveal differences in urinary concentration of dibasic amino acids when compared with normal dogs. However, through a recessive mode of inheritance, these dogs are able to transmit the disease to their offspring. Early detection of mutation carriers through cost-effective reliable methods is therefore essential for the implementation of breeding methods aimed at the eradication of the disease. Denaturing high-performance liquid chromatography (DHPLC) is a recently developed technique for rapid and efficient screening of nucleotide polymorphisms in polymerase chain reaction-amplified products. This technique was used for the identification of the C663T Slc3a1 mutation in Portuguese Newfoundland dogs. Polymerase chain reaction products amplified from a region containing the C663T locus were subjected to DHPLC analysis, and results were double checked by DNA sequencing. Results showed the presence of the mutation in 6 of the 22 dogs tested. Urine biochemical parameters correlated well with the number of mutated Slc3a1 copies, and homozygotes for the C663T mutation were the only dogs diagnosed with cystinuria. Sequence analysis confirmed the DHPLC results, demonstrating that the technique could be a reliable alternative to sequencing for the rapid and cost-effective identification of mutations in canine breeds.
